Traditionally, "Now" is an adverb. But I would say it's a preposition here because it can be modified by 'right' - - "There is no school in Myanmar right now". I can't think of a circumstance where "now" is used as a postpositive adjective.
Now is defined as at the present time or immediately. An example of now used as an adverb is the phrase right now, meaning someone is immediately going outside.
Of course now simply means the present moment, so by now means "a period of time ending at the present moment." So no, it is not right to say By now, I won't go to Melbourne and I am not sure what you mean by that. You can put by now at the beginning or at the end of a sentence.
